That depends on what mail server software you use.

Like the previous poster said, it depends on what email system your domain is using – most of the time, you go to the administration panel and click add new user.
If you're using cPanel, you click add / remove email accounts.
Realize also, however, that some hosts do not let you add additional email addresses – in which case your only alternative is to use a different mail service. Try Google for your Domain (search for it) it's free for up to 50 users.
